Directions of Trade Flows and Labor Movements between High- And Low-Population Growth Countries: An Overlapping Generations General Equilibrium Analysis
This paper considers a two-country world where the population in one country grows faster than the other, and investigates the implications of the addition of non-stationary population dynamics to a simple 2- commodity, 2-factor model of international trade within an overlapping- generations framework. The two countries in the world considered are assumed to be identical in every respect except, for their population growth rates initially. The effects of differential speed of population growth on relative factor endowments and patterns of international trade are explored by comparing simulation results obtained from the overlapping-generations general equilibrium model under autarky and trade scenarios. Unequal population growth rates are shown to give rise to differentials in wage rates and rentals for capital under autarky conditions. This, in turn, causes costs of production and relative prices to differ, creating the grounds for trade in the sense of Heckscher-Ohlin (HO). Yet, the results from simulation exercises indicate that static welfare results from the standard 2x2x2 HO model can not be generalized to hold in a dynamic setting with overlapping generations of individuals.Unequal population growth rates, labor migration, international trade, overlapping-generations
NAPO as a novel marker for apoptosis
Apoptosis or programmed cell death plays a pivotal role in embryonic development and maintenance of homeostasis. It is also involved in the etiology of pathophysiological conditions such as cancer, neurodegenerative, autoimmune, infectious, and heart diseases. Consequently, the study of apoptosis is now at center of both basic and clinical research applications. Therefore, sensitive and simple apoptosis detection techniques are required. Here we describe a monoclonal antibodyâdefined novel antigen, namely NAPO (negative in apoptosis), which is specifically lost during apoptosis. The anti-NAPO antibody recognizes two nuclear polypeptides of 60 and 70 kD. The antigen is maintained in quiescent and senescent cells, as well as in different phases of the cell cycle, including mitosis. Thus, immunodetection of NAPO antigen provides a specific, sensitive, and easy method for differential identification of apoptotic and nonapoptotic cells

It is assumed that one in ten women will develop breast cancer till the age 80. Heredity is the major risk determinant in breast cancer. 30% of the women with breast cancer have a positive family history and 10% of women is defined to have high risk of early onset breast cancer who have multiple family members of breast and ovarian cancer. A novel breast cancer susceptibility gene, BRCA2 was confirmed to account more than 40% of the mutations in familial breast cancer patients. BRCA2 tumor supressor gene encodes a 3418 amino acid protein with little or no homology to other known proteins. There is no significant data concerning the cellular functions of BRCA2 and many facts about BRCA2 are waiting to be uncovered. The gaps in the knowledge about BRCA2 must be filled by generating new points of views and technical approaches. Such studies require the cloning of BRCA2 and the presence of purified protein products. In this study we cloned a fragment of exon 11 and exons 19 to 27 as 2 overlapping fragments. Since more than 80% of the mutations result in the loss of the C-terminal of the protein and produce cancer phenotype, we expressed and purified the extreme C-terminus of the BRCA2 protein.Sayan, EmreM.S
MicroRNA Control of Invasion and Metastasis Pathways
Despite recent advances, cancer remains a leading cause of death worldwide. In developed countries, the incidence of colorectal and breast cancer has been stable, but no improvement in prognosis has been observed if the patient presents with metastases at diagnosis. This fact highlights the importance of therapeutic approaches targeting cellular invasion and metastasis programs as the next step in cancer treatment. During carcinoma progression a process called epithelialâmesenchymal transition (EMT) results in enhanced invasion and motility which is directly linked with loss of epithelial polarity and epithelial junctions, migration permissive cytoskeleton alterations, and the acquisition of mesenchymal properties. The recent discovery of microRNAs (miRNAs) controlling key cellular pathways has opened a new era in understanding how EMT pathways are modulated. In this review, we classify EMT regulating proteins according to their cellular localization (membrane, cytoplasmic, and nuclear), and summarize the current knowledge on how they are controlled by miRNAs and propose potential miRNAs for the transcripts that may control their expression

Bu çalıĆmanın amacı, ĂŒlkemizde özellikle yaygın din eÄitim-öÄretimi baÄlamında önemli bir misyonu olan Diyanet Ä°Ćleri BaĆkanlıÄıânın yeni medya karĆısındaki durumunu, merkez ve taĆra teĆkilatlarında (81 il mĂŒftĂŒlĂŒÄĂŒ), kamuoyuna din hizmeti götĂŒrme konusunda görevini ifa ederken, hangi sosyal medya mecralarını kullandıÄını nicel ve nitel kapsamıyla tespit etmektir. Ăzellikle 2019 Aralık ayında yaĆanan Covid-19 pandemisi sonrası dĂŒnya genelindeki dijital dönĂŒĆĂŒme paralel, ĂŒlkemizde de iletiĆim kanallarındaki deÄiĆimin Diyanet nezdinde nasıl gerçekleĆtiÄi ayrıca araĆtırmanın sorularındandır. Kurumun ve birimlerinin resmĂź web siteleri incelenerek hangi sosyal medya mecralarını kullandıkları tespit edilmiĆ, bu mecralarda öne çıkan birimler ve Facebook ve Twitter paylaĆımları içerik analizine tabi tutulmuĆtur. Bu çalıĆmanın amacı, yĂŒksek din öÄrenim gören öÄrencilerin öÄrenme iklimi algıları ile akademik özyeterlik ve akademik baĆarıları arasındaki iliĆkiyi incelemektir. Bu çerçevede dört temel araĆtırma sorusu cevaplanmaya çalıĆılmıĆtır: (i) YĂŒksek din öÄrenimi öÄrencilerinin öÄrenme iklimi ve akademik özyeterlik algıları ne dĂŒzeydedir? (ii) YĂŒksek din öÄrenimi öÄrencilerinin öÄrenme iklimi ve akademik özyeterlik algıları arasında anlamlı iliĆki var mıdır? (iii) YĂŒksek din öÄrenimi öÄrencilerinin yaĆ, öÄrenme iklimi ve akademik özyeterlik inançları akademik baĆarılarının anlamlı yordayıcısı mıdır? (iv) YĂŒksek din öÄrenimi öÄrencilerinin öÄrenme iklimine dair olumsuz algılarına iliĆkin görĂŒĆleri nelerdir? Karma yöntem ve yakınsak paralel araĆtırma deseni ile desenlenen araĆtırmada çeĆitleme stratejisi kullanılmıĆtır. AraĆtırmada, TĂŒrkiye'de devlet ĂŒniversiteleri bĂŒnyesindeki muhtelif Ä°lahiyat ve Ä°slami Ä°limler FakĂŒltelerinde yĂŒksek din öÄrenimi gören, altı fakĂŒlteden, nicel bölĂŒmde 1147 öÄrenciye Owen ve Froman tarafından geliĆtirilen, Ekici tarafından TĂŒrkçeâye uyarlaması yapılan Akademik Ăzyeterlik ĂlçeÄi ve Terzi tarafından geliĆtirilen, Ăniversite ĂÄrencilerine Yönelik Okul Ä°klimi ĂlçeÄi uygulanırken, nitel bölĂŒmde ise aynı fakĂŒltelerden 18 öÄrenciye görĂŒĆme formu doldurtulmuĆtur. 2019 Mayıs ayında gerçekleĆtirilen araĆtırma sonrası elde edilen nicel veri setine normallik testi uygulanmıĆ, verilerin analizinde; BaÄımsız Gruplar t Testi, Mann Whitney U Testi, Levene testi, Tek YönlĂŒ Varyans Analizi (ANOVA), Scheffe Testi, Kruskal Wallis H Testi, Pearson product-Moment Korelasyonu, Polyserial Korelasyonu ve Yol Analizi yapılmıĆtır. Nitel veriler betimsel analizle çözĂŒmlenmiĆtir. AraĆtırma sonuçlarına göre, yĂŒksek din öÄrenimi öÄrencilerinin öÄrenme iklimi algıları olumsuz çıkmıĆtır. YĂŒksek din öÄrenimi öÄrencilerinin genel olarak Akademik Ăzyeterlik algıları da olumsuz çıkmıĆtır. Okul Ä°klimi ĂlçeÄinin genel ortalaması ile Akademik Ăzyeterlik Algısı ĂlçeÄinin genel ortalaması arasında korelasyon orta dĂŒzeydedir. Loss of the branched-chain amino acid transporter CD98hc alters the development of colonic macrophages in mice
Comprehensive development is critical for gut macrophages being essential for the intestinal immune system. However, the underlying mechanisms of macrophage development in the colon remain elusive. To investigate the function of branched-chain amino acids in the development of gut macrophages, an inducible knock-out mouse model for the branched-chain amino acid transporter CD98hc in CX3CR1+ macrophages was generated. The relatively selective deletion of CD98hc in macrophage populations leads to attenuated severity of chemically-induced colitis that we assessed by clinical, endoscopic, and histological scoring. Single-cell RNA sequencing of colonic lamina propria macrophages revealed that conditional deletion of CD98hc alters the "monocyte waterfall"-development to MHC II+ macrophages. The change in the macrophage development after deletion of CD98hc is associated with increased apoptotic gene expression. Our results show that CD98hc deletion changes the development of colonic macrophages
ATM Regulates Differentiation of Myofibroblastic Cancer-Associated Fibroblasts and Can Be Targeted to Overcome Immunotherapy Resistance
Myofibroblastic cancer-associated fibroblast (myoCAF)-rich tumors generally contain few T cells and respond poorly to immune-checkpoint blockade. Although myoCAFs are associated with poor outcome in most solid tumors, the molecular mechanisms regulating myoCAF accumulation remain unclear, limiting the potential for therapeutic intervention. Here, we identify ataxia-telangiectasia mutated (ATM) as a central regulator of the myoCAF phenotype. Differentiating myofibroblasts in vitro and myoCAFs cultured ex vivo display activated ATM signaling, and targeting ATM genetically or pharmacologically could suppress and reverse differentiation. ATM activation was regulated by the reactive oxygen species-producing enzyme NOX4, both through DNA damage and increased oxidative stress. Targeting fibroblast ATM in vivo suppressed myoCAF-rich tumor growth, promoted intratumoral CD8 T-cell infiltration, and potentiated the response to anti-PD-1 blockade and antitumor vaccination. This work identifies a novel pathway regulating myoCAF differentiation and provides a rationale for using ATM inhibitors to overcome CAF-mediated immunotherapy resistance.SignificanceATM signaling supports the differentiation of myoCAFs to suppress T-cell infiltration and antitumor immunity, supporting the potential clinical use of ATM inhibitors in combination with checkpoint inhibition in myoCAF-rich, immune-cold tumors
Molecular mechanisms of cell death: recommendations of the Nomenclature Committee on Cell Death 2018.
Over the past decade, the Nomenclature Committee on Cell Death (NCCD) has formulated guidelines for the definition and interpretation of cell death from morphological, biochemical, and functional perspectives. Since the field continues to expand and novel mechanisms that orchestrate multiple cell death pathways are unveiled, we propose an updated classification of cell death subroutines focusing on mechanistic and essential (as opposed to correlative and dispensable) aspects of the process. As we provide molecularly oriented definitions of terms including intrinsic apoptosis, extrinsic apoptosis, mitochondrial permeability transition (MPT)-driven necrosis, necroptosis, ferroptosis, pyroptosis, parthanatos, entotic cell death, NETotic cell death, lysosome-dependent cell death, autophagy-dependent cell death, immunogenic cell death, cellular senescence, and mitotic catastrophe, we discuss the utility of neologisms that refer to highly specialized instances of these processes. The mission of the NCCD is to provide a widely accepted nomenclature on cell death in support of the continued development of the field
